The serine protease, DegP exhibits proteolytic and chaperone activities, essential for cellular protein quality control and normal cell development in eukaryotes. The P. falciparum DegP is essential for the parasite survival and required to combat the oscillating thermal stress conditions during the infection, protein quality checks and protein homeostasis in the extra-cytoplasmic compartments, thereby establishing it as a potential target for drug development against malaria. Previous studies have shown that diisopropyl fluorophosphate (DFP) and the peptide SPMFKGV inhibit E. coli DegP protease activity. To identify novel potential inhibitors specific to PfDegP allosteric and the catalytic binding sites, we performed a high throughput in silico screening using Malaria Box, Pathogen Box, Maybridge library, ChEMBL library and the library of FDA approved compounds. The screening helped identify five best binders that showed high affinity to PfDegP allosteric (T0873, T2823, T2801, RJC02337, CD00811) and the catalytic binding site (T0078L, T1524, T2328, BTB11534 and 552691). Further, molecular dynamics simulation analysis revealed RJC02337, BTB11534 as the best hits forming a stable complex. WaterMap and electrostatic complementarity were used to evaluate the novel bio-isosteric chemotypes of RJC02337, that led to the identification of 231 chemotypes that exhibited better binding affinity. Further analysis of the top 5 chemotypes, based on better binding affinity, revealed that the addition of electron donors like nitrogen and sulphur to the side chains of butanoate group are more favoured than the backbone of butanoate group. In a nutshell, the present study helps identify novel, potent and Plasmodium specific inhibitors, using high throughput in silico screening and bio-isosteric replacement, which may be experimentally validated.  相似文献

Cartilage replacement materials exhibiting a set of demanding properties such as high water content, high mechanical stiffness, low friction, and excellent biocompatibility are quite difficult to achieve. Here, poly(p-phenylene-2,6-benzobisoxazole) (PBO) nanofibers are combined with polyvinyl alcohol (PVA) to form a super-strong structure with a performance that surpasses the vast majority of previously existing hydrogels. PVA–PBO composites with water contents in the 59–76% range exhibit tensile and compressive moduli reaching 20.3 and 4.5 MPa, respectively, and a coefficient of friction below 0.08. Further, they are biocompatible and support the viability of chondrocytes for 1 week, with significant improvements in cell adhesion, proliferation, and differentiation compared to PVA. The new composites can be safely sterilized by steam heat or gamma radiation without compromising their integrity and overall performance. In addition, they show potential to be used as local delivery platforms for anti-inflammatory drugs. These attractive features make PVA–PBO composites highly competitive engineered materials with remarkable potential for use in the design of load-bearing tissues. Complementary work has also revealed that these composites will be interesting alternatives in other industrial fields where high thermal and mechanical resistance are essential requirements, or which can take advantage of the pH responsiveness functionality.  相似文献

An electrochemical approach to fabricate a nanostructured Fe/Pt-Fe catalyst through electrodepo-sition followed by galvanic replacement is presented. An Fe/Pt-Fe nanostructured electrode was prepared by deposition of Fe-Zn onto a Fe electrode surface, followed by replacement of the Zn by Pt at open-circuit potential in a Pt-containing alkaline solution. Scanning electron microscopy and energy-dispersive X-ray techniques reveal that the Fe/Pt-Fe electrode is porous and contains Pt. The electrocatalytic activity of the Fe/Pt-Fe electrode for oxidation of methanol was examined by cyclic voltammetry and chronoamperometry. The electrooxidation current on the Fe/Pt-Fe catalyst is much higher than that on flat Pt and smooth Fe catalysts. The onset potential and peak potential on the Fe/Pt-Fe catalyst are more negative than those on flat Pt and smooth Fe electrodes for methanol electrooxidation. All results show that this nanostructured Fe/Pt-Fe electrode is very attractive for integrated fuel cell applications in alkaline media.  相似文献

Microindentation hardness testing was applied to five types of highly-crosslinked ultrahigh molecular weight polyethylenes (UHMWPEs) for total joint replacements. UHMWPE's were crosslinked using the same total radiation dose (75 kGy; γ-radiation) either by the standard, single-step irradiation (one-step crosslinking) or by the newer, several-step irradiation (sequential crosslinking). Each irradiation step was followed by thermal treatment (annealing at 110 °C or remelting at 150 °C) in an inert atmosphere. We showed that: (i) the micromechanical properties were determined by the last thermal treatment step, while the number of irradiation cycles was insignificant and (ii) the values of microhardness, microcreep and microplasticity from the microindentation experiments were in excellent agreement with the changes of UHMWPE structure, characterized by IR and DSC. Statistical evaluation of the results, the agreement with theoretical predictions and the comparison with previous studies on similar systems demonstrated that microindentation was a reliable and sensitive method of UHMWPE characterization.  相似文献

老油田进入特高含水期后,剩余油分布更加零散,各种地质因素也因长期注水开发而发生了一定的改变.此时稳油控水难度进一步加大,为了进一步挖掘剩余油,控制含水上升速度、减缓产量递减,必须对注水井采取进一步的细分调整.基于水湿油藏注水井注水层段吸水效果,从注入水受力分析入手,运用达西定律推导出了水驱油藏水驱油全过程中的单位体积注入水沿竖直、地层两个方向运移的渗流速度公式,明晰了水驱油藏注水层段不同开发阶段砂岩吸水机理.研究结果表明,水驱油藏细分注水效果与岩石绝对渗透率、水相相对渗透率、注入水黏度、水动力压力梯度、注水井启动压力梯度、含水饱和度、地层倾角、油水密度差等参数有关.结合胜利油田断块油藏地质特征及细分注水工艺要求,制定了一套适合胜利油田不同断块区的细分注水技术政策界限:层段内渗透率变异系数小于0.3,砂岩条带宽度极比小于2.5,且东辛油区断块油藏层段内砂岩厚度小于6m,小层数小于4个;现河油区断块油藏层段内砂岩厚度小于5m,小层数小于5个;临盘油区断块油藏层段内砂岩厚度小于6m,小层数小于6个.矿场试验证实,该细分注水技术政策界限是合理、实用的,其为胜利断块油藏注上水、注够水、注好水,实现多层断块油藏的高效开发提供了技术保障,对类似油藏细分注水技术界限研究具有一定的借鉴意义.  相似文献

Urchin‐like Ag/ZnO hierarchical nano/microstructures have been synthesized through a facile low‐temperature hydrothermal growth method based on galvanic replacement mechanism. The experimental results show that the urchin‐like Ag/ZnO heterostructures are formed through the epitaxial growth of ZnO nanorods on the {111} facets of Ag nanoparticles along their own c‐axis. The photocatalytic properties of the products were evaluated by the degradation of RhB dye solution under ultraviolet irradiation, and the results show that the products exhibit significantly enhanced photocatalytic properties comparing with pure ZnO nanorods. The products with a Ag content of 35.64 atom % prepared with a Ag+ concentration in solution of 5 mM exhibit surprisingly high degradation rate (99.5%) for RhB dye solution (4 mg/L) after photocatalytic reaction for only 14 min under ultraviolet irradiation. The Schottky barrier formed at the metal‐semiconductor interfaces improves the segregation of charges and prevents the charge recombination, and thus significantly enhances the photocatalytic activities of the products. On the other hand, the high stability of the urchin‐like Ag/ZnO hierarchical nano/microstructures can effectively prevent the aggregation of nanostructures with simultaneously preserving high photocatalytic properties due to the existence of nanosized unites. An infection‐age virus dynamics model for human immunodeficiency virus (or hepatitis B virus) infections with saturation effects of infection rate and immune response is investigated in this paper. It is shown that the global dynamics of the model is completely determined by two critical values R 0, the basic reproductive number for viral infection, and R 1, the viral reproductive number at the immune‐free infection steady state (R 1<R 0). If R 0<1, the uninfected steady state E 0 is globally asymptotically stable; if R 0>1 > R 1, the immune‐free infected steady state E ? is globally asymptotically stable; while if R 1>1, the antibody immune infected steady state is globally asymptotically stable. Moreover, our results show that ignoring the saturation effects of antibody immune response or infection rate will result in an overestimate of the antibody immune reproductive number. Via utilizing the mixed‐ligand method, two novel Zn(II)‐containing meta‐organic frameworks with the chemical formula of {[Zn(L)(5‐HIP)]·H2O}n ( 1 ) and [Zn(L)(2,6‐NDC)]n ( 2 ) were prepared under the solvothermal conditions by applying aromatic dicarboxylic acids ligands (5‐H2HIP = 5‐hydroxyisophthalic acid; 2,6‐H2NDC = 2,6‐naphthalenedicarboxylic acid) and 1,4‐bis(benzimidazol‐1‐yl)‐2‐butylene (L). Due to its good water stability as well as the strong luminescent emission around room temperature, complex 2 has the high selectivity and sensibility of fluorescence detection to the ceftriaxone sodium (a kind of antibiotic) with the detection limit up to ppm lever. The treatment activity of the compounds on age‐related macular degeneration was assessed and the specific mechanism was investigated. First of all, the inflammasome activation in the endothelial cells of retina was evaluated with western blot. In addition to this, the down‐stream production of the inflammasome activation was also measured with ELISA detection kit.  相似文献
